The principle of spectrophotometer is that the continuous radiation light generated by a light source is decomposed into monochromatic light with a continuous wavelength by the dispersion system (monochromator), and the monochromatic light is irradiated on the sample cell, and absorption will occur when passing through the sample in the cell, by detecting the intensity S of the outgoing light and the intensity R of the incident light, the transmittance T = S/R. If the wavelength is continuously adjusted (i.e., scanning), the transmittance of the wavelength distribution in the biochemical sample can be obtained, and the transmittance with respect to the wavelength distribution is related to the composition and concentration of the sample.
